Nomarski microscopy

Nomarski microscopy is the same as DIC microscopy. To mount worms for microscopy:
- Place a drop of molten 4% Agar Noble onto a microscopy slide.
- Place a second slide over the first one in order to compress the Agar into a flat pad.
- Let Agar cool out.
- Remove top slide by sliding or lifting it off.
- The amount of buffer used is Voodoo. Some say 2.7 µl, other dont care, others make "oceans" on their slides. As a starting point place 4 µl of PBS or M9 on top of the Agar pad. You might use sodium azide to anesthesize the worms.
- Pick animals into the drop of buffer.
- Place coverslip on the Agar pad with buffer and worms.
- Place into the slide holder of a microscope.
- Do you remember how to do Köhler illumination?
- Don´t forget that we have objectives that need oil and others that must be used without oil.
- Clean objectives and condensor with a green Kimwipe after use.
