To do:
First read "Setting up crosses". Then pick 12 California(PS312) males on a spotted plate and let them get rid of all eggs and larvae that might stick to them. Cross them with J4's of Ppa unc 1 (make 4 plates with 3 hermaphrodites and 3 males each). Kill the males 24 hours later.
If this is your first contact with living animals, do the following (yes, this is trivial. or maybe not?):
Observe the F1:
- How many males do you see? Why are there males, after all?
- Are the F1 hermaphrodites wild type or unc? Both? Interpret the result!
- Isolate some wild type F1 virgins (J4 stage), if there are any. What happens in the F2?
More experienced worm breeders might try the following:
- Take some F1 males out of this cross and mate them to a dpy. Isolate 20 non-dpy F1'. What do you expect, what do you find? (The solution is here)
