DAPI staining

wash worms twice in PBS buffer (or M9)

afterwards fix them for 4 hrs with 4% formaldehyde

(alternatively fix them in Carnoy´s for 1,5hrs - overnight)

then wash them again twice in PBS buffer (or M9)

stain them afterwards with 1µg/ml DAPI either in the mounting solution already or in water

after staining overnight wash them again once in PBS

and then mount them in an antibleach mounting medium (e.g. vectashield or antifade)
