Carnoy´s

Carnoy´s is a good fixation for nuclei (alternatively to the formaldehyde fixation mentioned in DAPI staining)


wash worms twice in M9 then put them into Carnoy´s consisting of

3ml Ethanol, 1.5ml acetic acid, 0.5ml Chloroform

fix for 1.5hrs - overnight then rehydrate through 50% and 30% EtOH

stain with DAPI staining or Hoechst 33258
